Laboratory for Cellular Immunology and Diagnostics of Complement System and Syphilis

A Brief Description of the Laboratory

The investigations conducted by the Laboratory for Cellular Immunology and Diagnostics of Complement System and Syphilis cover the following three fields of immunology: determination of cellular immunity and natural resistance, measurement of complement system activation and deficiency, and diagnostics of some bacterial infections.

In the field of cellular immunity determination, sensibilization tests by antigens Mycobacterium tuberculosis (Quantiferon TB Gold) and in vitro lymphocyte activation tests with mytogens (PHA and ConA) and antigens (Borrelia burgdorferi s.l.) (TAL) are performed. The laboratory also carries out measurements of neutrophil granulocyte and monocyte activities in the light of phagocyting and intracellular killing potential and NK-cells activity with regard to extracellular killing of K562 targets.

In the field of measurement of complement system activation, the following laboratory analyses are performed: efficiency measurement of classical pathway (CH50), alternative pathway (APH50), and mannan pathway (in preparation), measurement of system activation by identifying waste products of the components C3 (C3dg) and C4 (C4d), determination of component concentrations of classical (C1q,r,s, C2, C3, C4, C5, C6, C7, C8, C9) and alternative pathways (FB, FD, FH, FI, FP), determination of C1inh regulator concentration and activity and determination of terminal lytic complex formation (C5b-9).

A large part of the laboratory work is focused on diagnostics of T. pallidum infection (syphilis). For diagnostic purposes, the following tests are performed: determination of the quantity of anti-cardiolipin (non-treponemal) antibodies (VDRL, RPR), determination of T. pallidum-specific (treponemal) antibodies by using TPHA, FTA, FTA-ABS, EIA methods and immunoblotting (INNO-LIA Syphilis Score), and identification of specific IgM antibodies against T. Pallidum with selective particle adsorption (19SIgM). The laboratory is also performing tests for the presence of antibodies against Helicobacter pylori and salmonella human pathogens (S. Typhi, Enteritidis, Parathypi A, B, C, Typhimurium and Newport).
